The treasure mission won't be offered again, so, without using cheats/exploits, you can only get into the Treasure Room twice (per playthrough). Once you turn in that mission, there is a second treasure mission that just repeats the last part of the story, starting at the lighthouse, and you can fight the leviathan and get back in the Treasure Room a second time that way. After you get into the Treasure Room the first time, you can go back to the lair again, but the door to the Treasure Room won't open.
